If you've jailbroken your iOS device and don't want to go through the hassle of manipulating the system files to make FaceTime calls over 3G then checkout jailbreak tweak FaceBreak.
Yaniv Dunan - developer of FaceBreak has been recently updated it to support iOS 5/iOS 5.0.1.
Here's a quick description of the jailbreak tweak on Cydia:
Enable FaceTime over 3G ! With 1 click and no setup, you can get FaceTime running over 3G and use it anywhere you want. No more looking up for nearby WiFi connection to use FaceTime. – Just install and Respring. FaceTime will now also work when on 3G.
The only downside of FaceBreak is that it costs $1.99 on Cydia while the manual method is free.
